Effect of Bi-1 Overexpression on Mitochondria in Human Fibrosarcoma Cells

|
The antiapoptotic protein Bax inhibitor-1 (Bi-1) was identified through a functional yeast screening designed to select for human cDNAs that inhibit Bax-induced apoptosis. Although Bi-1 has been known as a protein to have anti-apoptotic function against endoplasmic reticulum (ER) stress via regulation of Ca2+ levels between ER and cytosol, the role of Bi-1 in mitochondira was not clarified. In this study, we found that Bi-1 localized in both mitochondria and endoplasmic reticulum. Therefore, we investigated the effect of Bi-1 on the morphological alteration of mitochondria as well as ER using electron microscope (EM). Our data showed the morphological change of mitochondria and alteration of ER generation upon Bi-1 overexpression, implicating that Bi-1 might play an important role in mitochondria-mediated apoptosis. Taken together, we suggest that the study using electron microscopy might provide a new aspect in the therapy of diseases caused by abnormal apoptosis regulation.
